[Description]
CloverDirect TM tRNA Reagent for
Site-Directed Protein Labeling allows the pin-point labeling by
fluorescence dyes or biotin at a defined position of protein in
E.coli cell-free translation. The labeled
protein will be produced by just adding CloverDirect
TM reagent and the DNA template which is replaced at a
particular position with amber stop codon (UAG) to in vitro
translation system.
[Features]
Labeling position is decided by an amber codon which is the one of
stop codons. Pin-point labeling allows selection of a
labeling position which hardly interferes with protein structure and
function in comparison to other methods (e.g., chemical labeling and
GFP fusion production). Labeled aminoacyl-tRNAs are able to choose
from three fluorescent dyes (HiLyte Fluor 488, TAMRA, ATTO633) for
488 nm, 543 nm and 633 nm excitation lasers, respectively.
Pin-point fluorescence labeled proteins greatly
facilitate analyses in single molecule fluorescence analysis,
fluorescence energy transfer (FRET), cell imaging, and other
applications.
Pin-point biotin labeled proteins can be used for
efficient immobilization by biotin-avidin
interaction.
